Actigel T is used for the reversible coupling of thiol containing ligands and/or the purification of thiol-containing molecules. Appropriate ligands for immobilization to Actigel T include small molecules, peptides or proteins. During the reaction, a thiol-disulfide exchange takes place which attaches the ligand to the resin through a disulfide bond. This bond can then be cleaved by a reducing agent such as mercaptoethanol or dithiothreitol, which liberates the ligand.

This method is useful when the molecule of interest has a reactive thiol group, while other thiols in the sample are less active. The molecule of interest binds preferentially to Actigel T, leaving other components of the sample in solution. Unbound proteins are removed in the wash step after which bound protein/ligand is cleaved from the resin by a reducing agent.
