Blue Cellthru BigBead is a high-capacity dye-ligand resin for direct purification of enzymes and proteins from crude, or unclarified, feed streams such as serum, tissue extracts, cell cultures and fermentation harvests. The large bead size allows particulate material to flow between the beads while the soluble product is captured by the dye functional group. The flow properties of the beads allow the use of tall columns on scale-up. Blue Uniflow contains the blue dye, Cibacron blue 3GA, which has a high affinity for several enzymes, most notably the enzymes that bind a purine nucleotide. Some examples of enzymes that bind to Cibacron Blue F3G-A are phosphofructokinase, pyruvate kinase, and dehydrogenase. It is estimated that as many as 50% of enzymes are nucleotide-binding, so Blue-dyes are often used as one of many purification steps in a process. They also bind to several non-enzymatic proteins such as albumin, lipoproteins, and blood coagulation factors. Blue Cellthru BigBead is extremely stable in alkaline conditions and is compatible with most commonly used buffers. Most Cibacron Blue chromatography resins will leach blue dye over time, as is evidenced by the blue color of their storage buffer, reducing their capacity without even using it.
