Blue Uniflow contains the blue dye, Cibacron
blue 3GA, which has a high affinity for several enzymes, most notably
the enzymes that bind a purine nucleotide. Some examples of enzymes
that bind to Cibacron Blue F3G-A are phosphofructokinase, pyruvate
kinase, and dehydrogenase. It is estimated that as many as 50% of
enzymes are nucleotide-binding, so Blue Uniflow is often used as one
of many purification steps in a process. It also binds to several
non-enzymatic proteins such as albumin, lipoproteins, and blood coagulation
factors. Blue Uniflow is extremely stable in alkaline conditions and
is compatible with most commonly used buffers. Most Cibacron Blue
chromatography resins will leach blue dye over time, as is evidenced
by the blue color of their storage buffer, reducing their capacity
without even using it. Sterogene’s linkage chemistry ensures
that this effect is drastically minimized, ensuring stable binding
capacities over the life of the resin and virtually eliminating product
