Our CAP-8 IgG Purification Kit combines two conventional, non-affinity
purification methods to obtain IgG purity. The total IgG yield from
this kit is approximately 70%.
The new optimized precipitation conditions yield a granular precipitate
that is easy to remove by filtration alone. This simplifies scale-up
considerably. In addition, there is no need for clarification of
the starting serum, ascites, or egg yolk prior to purification since
the precipitation step also removes lipids and hydrophobic proteins.
From the filtrate, IgG is then purified on the SP Thruput Plus column.
The IgG is obtained with both high yield and purity.
This method was developed for IgG purification from serum, ascites
and egg yolk. The procedure delivers high yield, is reproducible
and easy to scale up. High-purity IgG is obtained in a one-step
cation exchange chromatography. Additional benefits are the delipidating
and viricidal effects of caprylic acid which simplify sample preparation
and facilitate process validation. The method has comparable performance
to affinity purifications at the fraction of the cost.
